Screening protein crystals for XRD data collection at synchrotrons

Abstract

To obtain a diffraction quality crystal it is often necessary to grow many crystals, mount, freeze, and then "shoot" them to see how well they diffract. Sometimes it may take 100s of crystal to find a good candidate. Transporting crystals to a beamline is tedious, but more importantly, beamtime at synchrotrons is expensive. Hence, using a synchrotron to screen crystals is not cost effective or efficient. To make better use of synchrotron time and resources we propose to begin screening protein crystals "at home" on a Bruker AXS Penteum 6000 X-ray diffractometer that exists at EMSL. Drs. Juers and Buchko request a couple days time on the instrument to explore the limits of the instrument using high quality lysozyme crystals, and if time permits, to "shoot" real crystals, such as DUF683.
