Longitudinal Proteomic Characterization of Human Cerebrospinal Fluid

Abstract
In order to facilitate future studies of diseased or drug-altered biological conditions by monitoring human cerebrospinal fluid (CSF) it is necessary to begin by creating a database/roadmap or masterlist of the normal human CSF proteome. We will use longitudinally-collected human CSF specimens from normal, healthy controls to cast a wide net that will capture the maximum number of potential candidate biomarkers in CSF that can then be augmented by characterizing CSF specimens obtained from relevant groups of patients. By focusing on the most relevant body fluid, cerebrospinal fluid (CSF), the closest fluid in contact with the brain (contiguous with the extracellular space of the brain cells), we will be able to directly characterize many of the proteins/peptides produced by the brain and central nervous system. In depth analysis of normal CSF will facilitate the future identification of novel CSF biomarkers that will have relevance to diseases and disease treatment, e.g., the compartmentalization of viruses and inefficient penetration of antiretroviral drugs into the brain; and that CSF rather than peripheral blood tends to be a better source of markers for certain types of diseases such as CNS AIDS.
