EPR characterization of hydrogenase

Abstract

Hydrogenase is a naturally occurring enzyme that efficiently converts H2 ->2H+ + 2e- and is reversible. Producing synthetic catalysts with these properties is highly desirable, but current analogs cannot match the low overpotentials and the fastest rates of the enzyme. Understanding how the enzyme works is an essential first step in the intelligent development of synthetic catalysts. Our fundamental goal is to understand the enzyme in order to develop synthetic catalysts, but fully characterizing our isolated protein to ensure that we do have active, properly folded hydrogenase is a necessary first step. This work proposes to characterize the EPR signature of the enzyme to ensure that we are working with the same material as other groups have, based on the published EPR spectra.
