Role of Human Intestinal Microbiota on Success of Surgical Weight Loss Procedures: Enhanced Understanding through application of NMR-based Metabolomics - Preliminary results

Abstract
We plan to use high-resolution 1H NMR spectroscopy to compare metabolite spectral profiles in aqueous extracts of human fecal material to obtain metabolic profiles significant to predicting outcomes of bariatric surgery. This data will be combined with existing project data to develop a coherent systems biology approach to understanding the metabolic changes in gut microbiota that may affect the outcomes of bariatric surgery. To address the need for preliminary data for an NIH supplementary grant submission due March 15th, we hope to gain a chance to produce preliminary data that will show sample tractability and data quality and perhaps identify a subset of metabolite peaks and their relative concentrations prior to submission of the supplementary grant request to NIH. Unlike the full study which would involve over 300-350 samples, we plan to examine 12 samples derived from 6 randomly selected samples in two different NMR tube sizes for evaluating the best conditions for NMR data collection. The samples derived from human subjects would fully comply with ASU's IRB (institutional review board) process and copies of approval and consent forms would be sent to PNNL's human subjects SME for approval prior to the samples being sent. We may continue to optimize experiments after the March 15th date, if instrument time is available. When the data collection is complete samples will be returned to ASU for further analysis and ultimate disposal.
