NMR profiling of monoclonal antibodies

Abstract

High resolution solution state NMR spectroscopy will be used to profile monoclonal antibodies. Two antibodies will be studied, 1) a candidate NIST standard Fab domain, and 2) trastuzumab, an FDA approved drug used to treat breast cancer. The NIST antibody is being profiled for a multilab comparability study to understand the potential value of NMR in antibody characterization. Trastuzumap is being characterized for a project aimed at developing a statistical method for characterization of macromolecular similarity, an application necessary for understanding how to approve generic versions of macromolecular pharmaceuticals ("biosimilars").
