Abstract

Single molecule fluorescence in situ hybridization (smFISH) is a powerful method to provide information about the spatial location, species, and activities of microbes through their gene expression patterns. Measuring mRNA copies in bacteria by smFISH has been demonstrated with only few examples due to the tediousness and complexity of the process that has not reached the maturity needed to serve general users. We will improve the smFISH process in sample preparation, instrumentation and data analysis to increase throughput and accuracy. The streamlined smFISH will be applied to a wide range of complex user samples.
