Tomography of Pseudarthrobacter chlorophenolicus

Abstract

Subcellular structures and macromolecular complexes can be revealed through cryo electron tomography of whole cells. Pseudarthrobacter chlorophenolicus is a soil bacterium which has novel metabolic pathways that may give insight into drought resistance mechanisms, in addition to being a non-infectious relative of a known pathogenic organism which may allow for study of infection mechanisms in a lower risk organism. Previous cryo electron tomography of this organism by our group has revealed several unique macromolecular structures of unknown function and origin. Collection of numerous tilt series with this proposal will help produce statistics on the incidence of these structures and their relationship to environmental growth conditions to gain a mechanistic understanding of their formation and function.
