Structural profiling of the molecular interactome to define phenotype

Abstract
The functional pathways that define phenotype are reflected by protein quinary structure; the transient encounters of proteins with other proteins, molecules, cofactors, and nucleic acids that are responsible for the molecular organization of a biological system. This project will develop and integrate three complementary technologies that leverage liquid chromatography-mass spectrometry (LC-MS) to measure protein quinary structure: (i) thermal proteome profiling (TPP), (ii) limited proteolysis (LiP), and (iii) fast photochemical oxidation of proteins (FPOP). Together, these techniques will provide comprehensive structural and functional data required for predicting and engineering biological phenotypes, the primary mission of the Predictive Phenomics Initiative.
