Pentosan Polysulfate Profiling with NMR Spectroscopy

Abstract

The proposed research will apply a heteronuclear nuclear magnetic resonance (NMR) spectroscopy method for profiling the molecular composition of pentosan polysulfate (PPS) in its pure active pharmaceutical ingredient (API, or drug substance) form. PPS is a synthetic glycosaminoglycan (GAG) derived from wood xylan (hemicellulose). Profiling PPS is important for establishing comparability among different preparations of PPS, for determination of physicochemical similarity. Insofar as PPS is a heterogenous molecular mixture, such profiling presents a challenging problem for which NMR is particularly well suited. (Note: The proposed project is being submitted specifically for data collection previously planned for a PNNL instrument that was shut down due to the global liquid helium shortage of 2022).
