Structure of the binding interface between tropomodulin and tropomyosin

Abstract

In striated muscles, actin thin filament architecture is critical for efficient contractile activity, and alterations in the thin filament length are linked to severe and often lethal muscle diseases. In this proposal, we will use NMR to determine the 3D structure of the interface between tropomodulin and tropomyosin, two actin-binding proteins that control length of thin filaments. Structural information on the binding interface between tropomodulin and tropomyosin will allow us to elucidate the molecular mechanism of tropomodulin binding to the thin filaments with respect to the thin filament length regulation.
