Project Title: Protein Structure Without Purification

Abstract

Purification remains a major technical and temporal bottleneck for protein annotation especially for membrane, filamentous and heterocomplex assembling proteins. If a target protein in a mixed population could be analyzed without purification, it would would increase throughput, which is a focus area in EMSL’s Bio-Atomic Imaging Research Area Roadmap and accelerate DOE biodesign efforts. Here we posit that cell-free lysates can empower rapid target protein structural analysis without purification. We will create a modified Build-and-Retrieve (BaR) method to solve structures of traditionally difficult samples including 3 BER relevant proteins. Success would commission a new capability and have direct applicability not only to multiple current EMSL user projects, but future user projects as well. We expect at least a 4x improvement in throughput due to the potential time savings.
