Algal Productivity Enhancements by Rapid Screening and Selection of Improved Biomass and Lipid Producing Phototrophs (APEX)

Abstract

This project uses a multi-pronged approach to improve oil content in algae capable of sustaining high areal biomass productivities in outdoor ponds. This includes mutagenesis techniques (EMS, ARTP) followed by chemostat selection and cell sorting (FACS) to isolate higher oil accumulating strains of two species, the diatom Nitzschia inconspicua, and algae Nannochloropsis gaditana. Other approaches in this proposal include genome engineering by CRISPR/Cas9, and bioprospecting with subsequent environmental selection under high temperature, high bicarbonate, high pH media conditions. We plan to use EMSL resources to isolate and characterize high lipid production strains of microalgae.
