Abstract
The purpose of this proposal is to develop portable electronic and optical biochip devices integrated with a novel nanoparticle labels for fast, low-cost, specific and sensitive detection of protein biomarker samples, which come from the pathogen-host cell 3-D system in the ongoing biomarker discovery projects of Response to Zoonotic Agents focus area in EBI. Apoferritin consists of a spherical protein shell composed 24 subunits surrounding an aqueous cavity with a diameter of about 8 nm capable of accommodating around thousands small molecules. In this proposal, numerous small molecules(electroactive or fluorescence marker) will be entrapped into the cavity of apoferritin to form apoferritin nanoparticle label for high sensitive electronic and optical biosensing protein biomarkers. The surface of the small molecules entrapped apoferritin will be functionalized with DNA probe or protein antibody. SEM, TEM , AFM , will be used to characterize the entrapped marker in apoferritin cavity, as well as the apoferritin nanoparticle.Infrared Spectroscopy will be used to indentify the modification steps. Fluorescence spectrum and UV spectrum will be used to measure the activity of conjugated DNA or protein on the surface of apoferitin nanoparticle, and intensitiy of fluorescence markers.Project Details
