Characterization of Novel Charge Ladders by Capillary Electrophoresis-FTICR Mass Spectrometry

Abstract
We believe that CE-ESI-FTICR-MS will be important in the characterization of complex charge ladders of proteins and aminoglycosides. The reaction of Lys- -NH2 groups of bovine carbonic anhydrase II (BCA II) with iodoacetic anhydride results in an iodoacetamide charge ladder. The proteins which constitute this charge ladder can be further modified by reaction with a thiolate nucleophile (neutral, negative and positively charged thiol, Scheme 1) or a tertiary amine (quinuclidine, Scheme 1). Several such protein derivatives were formed, but they can not be sufficiently characterized by CE alone; CE-ESI-FTICR-MS would therefore be extremely useful to characterize these derivatives. We are also enclosing a sample of a phenylglyoxal charge ladder of lysozyme. We are interested in determining by CE-ESI-FTICR-MS whether the derivatives which are formed by reaction of the Arg residues of lysozyme with phyenylglyoxal represent a 1:1 or 2:1 adduct with phenylglyoxal. This charge ladder must be analyzed on a polybrene coated capillary.
