Characterization of Post-Translational Modification of the A-Subunit from Heat-Labile Enterotoxin using FTICR Mass Spectrometry

Abstract

Project description: The goal is to use the high mass measurement accuracy provided by FTICR MS to characterize posttranslational modifications of a 26 kDa protein. This protein is the A-subunit of heat-labile enterotoxin, an AB5 toxin from E.coli, associated with cholera-like diarrhea. The A-subunit is an ADP-ribosyl transferase which is capable of self-modification on arginine residues, upon addition of NAD. We are trying to determine how many residues are modified and locate the sites of modification.
