Determination of the proteome of drosophila lipid droplets

Abstract

My research goal is to understand the way molecular motors such as kinesin and dynein move cargos in vivo. To investigate this, I have developed a model system, the bi-directional transport of lipid droplets along microtubules, in early Drosophila embryos. I am a biophysicst by training, and have developed biophysical tools to quantify the motion of individual lipid droplets. In order to better understand this process, I would like to collaborate with scientists at Pacific Northwest national laboratory, to determine the proteome of the lipid droplets. In my lab we would purify the proteins bound to the droplets, and then their identity would be determined using PNL's mass-spectroscopy equipment. Overall, the goal is FUNCTIONAL PROTEOMICS--purify the droplets, determine the proteins bound to them, and then use my biophysical functional assays combined with the power of drosophila genetics, to further understand what the identified proteins do.
