Towards Better Understanding of the Siderophore Uptake Process in Escherichia coli

Abstract

Using siderophores to scavenge metal ions from the host is critical to the thrive of the bacteria during infections. The metal-chelated siderophores are reimported into the bacterial cells through a subgroup of ATP-binding cassette (ABC) importers. However, the detailed molecular mechanisms of these importers are not fully understood. In this proposal, we expect to study three main siderophore ABC importers from uropathogenic E. coli by cryo-EM, so to better understand their unique roles in bacterial growth. The knowledge gains here will be informative because it could be used to develop drugs to target this unique biological process in human pathogens.
