Cryo-EM structures of ion channels in calcium signaling

Abstract
The aims of this ongoing work are to determine 3D structures of ion channels involved in calcium signaling. These aims are central to our NIGMS R35 award GM13192, Mechanisms of Ion Channels in Calcium Signaling.The goal of the current proposal to is to further our progress in determining high-resolution cryo-EM structures of the eukaryotic ion channels Orai, and MCU, and BEST that represent different gating conformations of these channels. We have identified appropriate conditions for data collection and collected preliminary cryo-EM datasets. 2D classification and preliminary 3D structures from these data inspire confidence for continued progress. Successful completion of the proposed studies will define structural principles for ion permeation, ion selectivity, and gating by these channels. These mechanistic insights will be of interest to a broad scientific community and may lead to approaches to pharmacologically modulate the channels (e.g. for immune control, cystic fibrosis, and cancer).
