Structure of retroviral integrase-DNA complexes

Abstract
Integration of retroviral genome is mediated by viral integrase (IN) into host genome and is an essential step in retroviral replication. IN multimerizes on viral DNA to produce intasome and carry out catalytic steps. We are using cryo-EM to determine the structure of HIV-2 intasome in presence of a FDA approved HIV-1 IN inhibitor. The structure will fill an existing knowledge gap in the field in understanding the assembly mechanism of retroviral intasomes. HIV-2 is a significant pathogen and number of infected individuals is on rise. Due to continual development of drug-resistance, it is imperative to continually identify novel drug target against HIV-1 and HIV-2.Project Details
