MntR interactions with RNA Polymerase in the mneP Promoter Region

Abstract

The manganese transport regulator (MntR) of Bacillus subtilis has long been known to repress uptake of manganese by binding to its operator site and blocking access of RNA polymerase to the promoters controlling expression of manganese uptake proteins. More recently, it has been discovered that MntR binds to 3-4 sites upstream of genes encoding a manganese export protein and can activate expression of those proteins. We propose to investigate the mechanism of activation by obtaining the cryo-EM structure of MntR and RNA polymerase interacting on the mneP promoter sequence.
