Abstract

The next stage in structure determination of the BRCA1/BARD1 heterodimer complex requires the assignment of aliphatic side chain H-1 and C-13 resonances. Typically, HCCH-TOCSY experiments on fully prononated samples are used for this purpose. These spectra are complicated due to the large number of resonances and overlap of signals. Therefore, the sensitivity and resolution afforded by higher field instruments are required. We propose to collect an HCCH-TOCSY and an C(CO)NH-TOCSY with H-2 decoupling at 600 MHZ and C-13 edited and N-15 edited NOESY on fully protonated samples at 800 MHz.
