Visualization of Gamma H2AX Foci

Abstract

Genomically unstable cells have been demonstrated to release a "factor" into the medium which when transferred to stable cells results in 100% cell death. First demonstrated by Nagar and Morgan (2004), this factor has been termed DIE or the Death Inducing Effect. Prior to cell death, DIE media has been shown to produce double strand breaks in stable cells. Using the Gamma H2AX assay of double strand breaks, this proposal will look at the correlation between foci number and DIE factor concentration using media titrations. Additionally, a time course for the production and nuclear location of foci will be performed. Imagining facilities in EMSL will be used for visualization and image acquisition necessary for subsequent data analysis.
