NMR Studies of Nudix proteins from D.radiodurans

Abstract

We are in the process of cloning and expressing proteins from D.radiodurans that are involved in various aspects of DNA repair and maintaining DNA integrity. Among these some of the nucleotide diphosphate binding proteins (Nudix) hydrolyze damaged nucleoside triphosphates to prevent their incorporation into DNA. We will try to better understand the activities of the large number (19) of these proteins which occur in the radiation resistant bug D.radiodurans by carrying out structural studies. Most of the proteins are in the 150-200 amino acid size range, and the only homologs that have been solved to date had structures determined with NMR.
