Sensitive Detection of Protein Phosphorylation

Abstract

EHSD (FS) LDRDThe objective is to demonstrate the applicability of ICP/MS to the sensitive detection and quantitation of phosphorylation on peptides and proteins. The scope is to develop sufficient preliminary data to form the basis of a full project proposal to DOE OBER. The expected results are two-fold: 1)the demonstration of direct detection and quantitation by ICP/MS of less than a picogram of phosphorous attached to a protein or peptide and 2) the correlation of an LC-ICP/MS chromatogram and an LC-ESI/MS (or MALDI/TOF) chromatogram of a phosphoprotein. These would allow the determination of the degree of peptide phosphorylation and determination of the amino acid sites of phosphorylation respectively.Use of the high resolution ICP/MS will be required.
