Single-molecule imaging at LB bilayers

Abstract

The proposed experiments with the LB trough are to create lipid bi-layer membranes with low concentrations of either fluorescence labeled lipids and/or low concentrations of fluorescence labeled membrane peptides and proteins. We will prepare the membranes on glass cover slides which will be subsequently placed into a sample chamber to keep the membranes in a wet, controlled environment. The sample chamber mounts onto our inverted, fluorescence microscope, so that we can record the lateral and rotational diffusion (and possibly the conformational motions) of the fluorescence labeled substance in the lipid bi-layer membrane and should serve as control experiments for our single-molecule fluorescence experiments in cell membranes.
