Optimization of LC-MS-based metabolomics pipeline

Abstract

The demand for LC-MS untargeted metabolomics analyses within EMSL user program has substantially increased during the past few years. As user facility and leading institution in mass spectrometry technological development and applications, EMSL needs a robust high-throughput LC-MS-based metabolomic capability for rapid yet comprehensive analysis of large number of environmental/biological samples. The proposed capability development addresses this demand by optimizing existing LC-MS chromatography pipeline for untargeted metabolomics to enhance metabolite recovery, detection, and identification. Specifically, we propose to: i) increase metabolome coverage through access to polar metabolites, ii) increase throughput by optimizing chromatography, and iii) increase identification confidence by enabling refinement of the existing metabolite libraries using validated exact masses, retention times and tandem mass spectra (MS/MS).
