Determine 3d-structure of chemokine in apo form and in complex with its native receptor by solid state NMR

Abstract

Structural biology has significantly accelerated the development of drug leads in protein targets for which 3d structures could be obtained either by XRAY crystallography or by NMR spectroscopy. Unfortunately, elucidating 3d structures in the most important class of drug targets, the G protein coupled receptors, has proven to be most challenging. Recent, dramatic advances in high resolution solid state NMR spectroscopy have unlocked the feasibility of delineating 3D structures of GPCRs and to characterize their conformational dynamics in various states of activation.
