Isolation of Isotopically-Labeled Nucleic Acids

Abstract

The primary goal of this project is to develop and implement new approaches for using isotopic labels in NMR-based DNA structure determination. 13C and 15N-labeled RNA and DNA will be isolated from bacterial cells grown in a medium containing isotopically-labeled carbon and nitrogen substrates. The cells are lysed to yield total nucleic acids which necessitates the separation of deoxyribonucleotides from ribonucleotides, and subsequently the separation of the individual nucleotides and ribonucleotides. Isolation of the individual nucleotides is a multi-step process, the first of which is hydrolysis of the total nucleic acids from the lysate. The hydrolysis is monitored by reverse-phase HPLC.
