COVID-19: Structural study of RNA-dependent RNA polymerase of SARS-CoV-2

Abstract

The outbreak of COVID-19 by SARS-CoV-2 infection has induced a global health crisis with millions of infections and about 20 thousand deaths. The replication of SARS-CoV-2 is dependent on RNA-dependent RNA polymerase (nsp12) and its cofactors of other nsp proteins. Here we prepared the complexes of nsp12 with its cofactor proteins, RNA substrates, and small molecules. By determining structures of these complexes, we will not only provide mechanistic understanding on the replication mechanism of SARS-CoV-2 but also provide structural basis for drug design against this devastating disease.
