Structure and mechanism of neurotransmitter-gated ion channels

Abstract

The objectives of this research are to elucidate the structures of mammalian neurotransmitter receptors by single particle cryo-EM in multiple distinct conformational states in which the receptors are bound with selected agonists, antagonists, allosteric modulators and/or ion channel pore blockers. We will also elucidate the structures of neurotransmitter receptors in complexes with auxiliary proteins that modulate receptor activity. The proposed studies will focus on ionotropic glutamate receptors, pentameric Cys-loop receptors and proton activated acid-sensing ion channels (ASICs). These studies will not only reveal the overall architectures and structures of neurotransmitter receptors and their complexes with auxiliary proteins but they will also provide detailed, structure-based insight into their mechanisms of action.
