Advanced 21T FTICR MS instrumentation and methods for functional proteomics

Abstract
The 21T FTICR-MS at the EMSL provides extremely high mass accuracy and resolution enabling superior depth of characterization for natural organic matter and intact mass measurement of denatured proteins.1 However, the technology utilized in the frontend of the spectrometer for the 21T FTICR-MS is dated and severely limits capabilities for the top-down characterization of proteins and native protein complexes. The proposed work builds upon EMSL’s leadership in proteomics and will yield a world-class platform with unmatched capabilities for structural biology and top-down characterization of the functional proteome. We will continue development of a hybrid Orbitrap-FTICR-MS with advanced tandem mass spectrometry (MS/MS) capabilities and data acquisition strategies to enable precise proteoform2 characterization and the molecular machines3 the proteoforms assemble into natively. The developed capabilities will be used to characterize native protein-metabolite complexes as a proof of principle for protein function and biological pathway elucidation in plants. All development work will be performed using the 12T magnet in EMSL 1621 and will not interfere with normal operation of the 21T FTICR-MS. Although this proposal is focused on top-down proteomics and native protein complexes, all instrumentation and method development will enhance small molecule/complex mixture analysis as well.
