Improving on MALDI-MS imaging of Environmental Microcystins

Abstract
This proposal aims to improve the spatial detection of microcystins (MC) in liver tissue of mice gavaged with most common MC congener, MC-LR. MCs are common environmental contaminants, cyclic heptapeptides, which have a main impact on the liver due to their binding to protein phosphatases 1 and 2A. MCs appear in lakes and reservoirs during harmful algal blooms, pollute water and create hazard for its further consumption and recreational activities. Preliminary MALDI-MS imaging experiments performed at University of Toledo and Wayne State University have yielded ambiguous results in terms of MC-LR detection in liver tissues, mainly because non-automatized MALDI matrix deposition devices and a low-resolution mass spectrometer (MALDI-TOF) have been used. Considering that EMSL possesses more sophisticated instrumentation for MALDI matrix deposition (HTX-TM Sprayer) and MALDI-MS imaging (15T-MALDI-FTICR), this project aims to provide unambiguous information about deposition and metabolic fate of MCs in the liver.
