High throughput generation of small molecule probes for discovery of unique cellular functions using Substrate Activity Screening

Abstract

We propose a method to demystify the massive catalog of proteins-of-unknown-function accumulated by recent advances in genome sequencing by direct assignment of function. We will query enzyme activity using large libraries of non-natural fluorogenic substrates to create an easy, high throughput means of screening proteins for unique functions. This project supports the EMSD/EMSL goal of establishment of a Digital Phenome through providing a method to create rich functional profiling data. The simplicity of the solid-phase synthesis approach will allow EMSL to create large, customizable, small molecule probe libraries with undergraduate-level chemical synthetic expertise.
