Structural identification of novel, cancer associated metabolites deriving from cysteine metabolism

Abstract

Activation of the oncogenic transcription factor NRF2 in cancer cells is associated with metabolic changes that remain poorly understood. My research group has identified that NRF2 activated cancers display an accumulation of novel metabolic fates of cysteine, that we have characterized as deriving from spontaneous reactions between cysteine and endogenous metabolites. While my lab has been able to verify key aspects of their formation and chemistry, we do not yet have unambiguous verification of their chemical structures. Our goal is to have the Cryo-EM group use their MicroED protocol to address this knowledge gap by measuring the structure of our generated standards.
