Cryo-EM Comparison Between Cryo-FIB-EXLO and Cryo-FIB On-The-Grid Prepared Yarrowia Lipolytica Yeast

Abstract

Our DOE funded research (DE-SC0020511) has enabled us to develop cryo-sample handling methods that eliminate the use of a cryo focused ion beam (cryo-FIB) instrument for specimen manipulation. By incorporating cryo ex situ lift out (cryo-EXLO), cryo handling of FIB specimens are performed with micromanipulation apparatus outside the FIB. Cryo EXLO improves speed, throughput, and reproducibility of cryo-FIB specimens, lowering the cost per specimen, and potentially leading to enhanced scientific discovery. Previous cryo-EXLO work was performed on plunge-frozen yeast. In this research, we expand our cryo-EXLO methods to high pressure frozen (HPF) prepared Yarrowia Lipolytica which is considered a model yeast for biofuel, biotechnology, and environmental applications. Applying cryo-EXLO to HPF vitreous samples will enable a larger variety of sample types to be manipulated since thicker arrays of cells or tissue may not be amenable to plunge freezing.
