Structure of the PR domain of RIZ1 tumor suppressor

Abstract
RIZ1 is a tumor suppressor with histone lysine methyltransferase activity that provides a link between dietary and genetic causes of cancer. The enzymatic activity is conferred to RIZ1 by its PR domain. We plan to determine the structure of the PR domain from RIZ1, using standard double and triple resonance NMR experiments. We have been awarded six weeks of 600 and 750 MHz time at PNNL in the current cycle (February-March 2004) for two different RIZ constructs. We request additional three weeks of NMR time on the 600 MHz spectrometer equipped with a cold probe to characterize the longer RIZ1 fragment that can be prepared only in very limited quantities.
