Proteomic Analysis of Shewanella oneidensis Biofilms

Abstract

Our research is directed to elucidate the molecular mechanism of biofilm formation in Shewanella oneidensis. This microbe is a geochemically important Fe(III)- and Mn(IV)-reducing microorganism, which can also use other metals as electron acceptor, including U and Cr. DOE is planning to use this microbe for bioremediation field projects to remediate some of its heavy metal contaminated superfund sites. Critical for the microorganism's performance in nature but also in those engineeed settings is its close association with metal oxide surfaces, specifically with Fe- and Mn-oxides that are commen in soil and sediments.

In the proposed experiments we seek a proteomic analysis of this microbe and of mutant strains when forming a biofilm. The genome of S. oneidensis is sequenced and the group of Dr. Richard Smith and Dr. Mary Lipton as Senior Research Scientist have developed a mass tag database of this microbe. This allows to examine the presence to the protein profile of this microbe. We wish to examine the protein profile of biofilm cells and selected mutants. We expect that we have about 10 samples that we wish to have analyzed initially. Results from these experiments would tremendously enhance our NSF-funded research and would result in a publication with your PNNL partners as collaborators.
