Abstract

We have synthesized several 15N,13C-labeled oligomers (labeled residues in bold) 5'-GGGAACAACTAG-3':3'-CCCTTGTTGATC-5' and 5'-GGGATCCACTAG-3':3'-CCCTAGGTGATC-5' which contain 8-oxoguanine (underlined), a stable product of oxidative DNA damage. 8-Oxoguanine is a substrate for the base excision repair (BER) DNA glycosylase enzymes which recognize and remove the damaged base. In this proposal we wish to study the structure and dynamics of the damaged DNA in different sequence contexts to identify the properties at the damaged site which are responsible for recognition by the DNA glycosylases. We have also synthesized 15N,13C-labeled 5'- CTCTTTXTTTCTC-3' :3'-GAGAAACAAAGAG -5' that contains an abasic site and binds to the BER enzyme, Formamidopyrimidine-DNA glycosylase (Fpg). Fpg acts as an apurinic/apyrimidinic lyase and we hope to gain further knowledge of this activity by studying structure and dynamics of the abasic DNA.A more detailed description of the project is available from the MS&D administrative staff.
