Molecular analysis of exRNA carrier subclasses

Abstract

As a consortium site, PNNL will perform proteomic and lipidomic analysis of exosome RNA (exRNA) carrier subclass samples generated using differential centrifugation, density gradient centrifugation, size exclusion chromatography, and immunomagnetic separation. Proteins and lipids from the exRNA carrier subclass samples isolated by the other sites will be individually extracted, digested (in the case of proteomics analysis), cleaned up, and analyzed by LC-MS/MS using high-resolution hybrid Orbitrap MS instruments at PNNL under optimized conditions for proteomics and lipidomics analysis, respectively. The resulting MS data will be analyzed using well-established data analysis pipelines at PNNL for protein and lipid identification and quantification. The final deliverables for this project will be the protein and lipid quantitation results in the format of plain text files or spreadsheets. The raw mass spectrometry data can also be provided, if requested.
